Oh yes, and remember to edit the actual podcast afterwards. Not everything needs to be kept in. I don't just mean all the awkward pauses, I mean the uninteresting tangents and alternate takes of flubbed lines. Make the panel look more professional than they actually are!
I imagine it'd be a really good idea to have a practice run first, though. Talk about any old rubbish (not even anything related to game devving, just any topic you can form a few questions and discussion points around), and get to grips with how each other behaves and how the format would work.
I'm near constantly listening to podcasts on my way into work, so a podcast like that would be really fun to listen to!

Game design is best debated, I think. Trying to lecture on it solo doesn't appeal.

I guess chronologically you'd start with the inro. Having an idea, and script ready, laying out your plans, etc.
